Abstract
As for any new drug, developers of oligonucleotide (ON) therapeutics need to be questioning their toxicity potential or the risk of causing unwanted toxicities or side effects. This chapter focuses on hybridization-independent toxicities of the most abundantly characterized ON class, antisense oligonucleotides. It will review the potential mechanisms underlying hybridization-independent toxicities and describe some specific concerns for the development of this therapeutic class. Finally, a strategy to address hybridization-independent toxicities will be presented.
