Les sels contenant des cations organiques sont une classe importante de composés cliniquement utilisés. Ils possèdent de nombreuses propriétés qui sont utilisées pour des applications biologiques, dont, entre autres, la possibilité de perturber les membranes cellulaires. Dans cette thèse, nous nous intéressons particulièrement au débalancement de l’homéostasie induit par l’insertion de certains sels d’imidazolium dans la membrane phospholipidique. La perturbation membranaire qui en résulte ouvre la porte au développement de certaines applications potentielles, telles que le traitement de maladies provenant des dysfonctions du transport ionique transmembranaire, ou bien comme stratégie émergente pour attaquer efficacement des microorganismes indésirables, comme les bactéries multirésistantes, fléau des temps modernes. Pour ce faire, nous avons démontré le transport transmembranaire d’anions effectué par une nouvelle classe de transporteurs mobiles organiques : des sels d’imidazolium fonctionnalisés par des groupements adamantyles. Le contrôle du processus de transport peut être réalisé par l’ajout de cyclodextrines, qui conduit à la formation d’une entité supramoléculaire incapable de perturber la membrane phospholipidique. L’ajout d’un composé invité compétitif permet de disloquer le transporteur du complexe supramoléculaire et de restaurer l’activité de transport transmembranaire. Ce système a ensuite été étudié pour une application potentielle comme stratégie d’administration de médicament contrôlable. La toxicité des sels d’imidazolium contre des bactéries Bacillus thuringiensis a été démontrée comme étant réversible, selon le même modèle que le transport transmembranaire. Nous présentons également un essai spectroscopique simple qui nous permet de quantifier la perméabilisation des membranes des bactéries Escherichia coli. Cette nouvelle approche permet le suivi de la perméabilisation de la membrane bactérienne, induite par le chlorure de benzalkonium ou par d’autres composés cationiques.
