La production des mouvements chez l’humain, et également chez plusieurs autres mammifères, s’exécute naturellement lors de la réalisation d’une tâche motrice complexe. Ces mouvements hautement coordonnés nécessitent l’intégration de plusieurs commandes provenant de différentes régions du cerveau, notamment les aires motrices du cortex cérébral, les ganglions de la base et le cervelet. Malgré les progrès récents en neurosciences, l’apprentissage de tâches motrices complexes n’est pas très bien compris. La manière dont le cerveau intègre et mémorise la séquence motrice reliée à une activité motrice n’est pas bien connue. La protéine kinase B (PKB/Akt) est sérine/thréonine kinase hautement conservée parmi les mammifères et régule plusieurs métabolismes cellulaires tels que la prolifération, la survie et la plasticité cellulaire. Akt possède trois isoformes (Akt1, Akt2 et Akt3) qui sont encodées par trois gènes distincts. Il est important de noter qu’Akt3 est significativement plus exprimée dans le cerveau que ses deux autres isoformes Akt1 et Akt2. Glycogen Synthase Kinase-3 (GSK-3) comporte deux isoformes, soit GSK-3 alpha et GSK-3 bêta. Akt et la protéine kinase GSK-3 sont intimement connectés. En effet, il est établi qu’Akt réduit l'activité de GSK-3alpha/bêta par sa phosphorylation sur la sérine 21/9 respectivement. Une perturbation de la voie de signalisation Akt/GSK-3 a été reliée à plusieurs pathologies telles que la dépression, la schizophrénie et les troubles bipolaires. Le coeur de mes recherches a été d'étudier la capacité d’apprentissage, de performance et de mémorisation d’une tâche motrice complexe chez des souris ayant une délétion génétique d’Akt3 (Akt3 KO). Les résultats sur le rotarod ont démontré un ralentissement de l’apprentissage moteur chez les souris Akt3 KO en comparaison aux souris sauvage (WT). De plus, les résultats obtenus démontrent un changement de la phosphorylation de GSK-3 alpha/bêta et ainsi suggèrent que le déficit d’apprentissage moteur serait causé par une hyperactivation de GSK-3 alpha/bêta, induite par la délétion génétique d’Akt3. Il est intéressant de noter qu’un traitement chronique au lithium, connu pour inhiber directement et indirectement la protéine GSK-3, a corrigé le déficit d’apprentissage des souris Akt3 KO. Nos résultats ont pu établir une implication importante de la voie Akt3/GSK-3 dans l’apprentissage moteur.
