Étude de l'implication neurologique et immunologique de la voie costimulatrice CD27/CD70 dans la sclérose en plaques

Abstract
Introduction : La sclérose en plaques (SEP) est une maladie inflammatoire chronique où des lymphocytes autoréactifs peuvent altérer l’intégrité de la barrière hématoencéphalique (BHE) et transmigrer dans le système nerveux central. La voie CD27/CD70 fournit un signal costimulatoire régulant la fonction effectrice des lymphocytes. Notre laboratoire a montré que l’expression de CD70 est plus élevée sur les lymphocytes des patients SEP comparativement aux sujets sains. Notre hypothèse est que la voie CD27/CD70 est centrale au phénotype pathogénique pro-inflammatoire et à la transmigration des lymphocytes T à travers la BHE dans la SEP. Méthode : L’étude de l’implication de la voie CD27/CD70 dans la transmigration lymphocytaire nécessite des cellules endothéliales de la BHE (CEBHE), un modèle in vitro de la BHE. Un essai de migration a évalué la capacité des cellules T polarisées en TH1/TH17 ou stimulées avec du TGF-β à traverser la BHE. L’expression de CD70 sur les CEBHE a aussi été examinée par cytométrie en flux, immunobuvardage de type Western et qPCR. De l’immunohistofluorescence a été effectuée sur des sections de cerveau de patients SEP afin d’y détecter des cellules CD27+/CD70+. Résultats : L’expression de CD70 sur les CEBHE est augmentée en condition de culture pro- inflammatoire. Les lymphocytes T CD70+ polarisés en TH1/TH17 maintiennent leur profil pro- inflammatoire après leur transmigration à travers la BHE. Des cellules immunitaires CD70+/CD27+ sont présentes dans les lésions du cerveau des patients SEP. Conclusion : La voie costimulatrice CD27/CD70 est impliquée dans la réponse inflammatoire dans le contexte de la SEP.
