La flocculosine est un glycolipide de 854 Da qui a été reconnu responsable de l'activité antagoniste de Pseudozyma flocculosa, un agent de lutte biologique contre le blanc et principe actif du biofongicide Sporodex®. Il a été démontré récemment que cette molécule avait une activité délétère contre plusieurs microorganismes. Or, selon le dernier rapport de l'organisation mondiale de la santé, les maladies infectieuses arrivent toujours au deuxième rang des principales causes de mortalité au monde. De plus, l'apparition croissante de microorganismes résistants aux agents antimicrobiens a considérablement réduit l'efficacité de l'arsenal thérapeutique disponible, ce qui accentue l'urgence du besoin de nouvelles molécules antimicrobiennes. L'objectif général de ce projet était donc d'évaluer le potentiel antimicrobien de la flocculosine afin de préciser son spectre d'activité, d'évaluer sa toxicité, de clarifier son mode d'action et de définir son implication dans l'écologie de P. flocculosa . En conditions acides, la flocculosine a démontré des effets délétères et très rapides contre une vaste gamme d'agents infectieux comprenant levures et bactéries. De plus, la molécule n'a pas été affectée par les divers mécanismes de résistance présents et l'association de la flocculosine avec l'amphotéricine B a révélé un fort synergisme, suggérant son utilisation comme adjuvant afin de réduire la toxicité et de contourner certaines résistances. Par ailleurs, la flocculosine n'a démontré aucune toxicité sur les différentes lignées cellulaires humaines exposées. Il a aussi été démontré que le glycolipide agissait au niveau de la membrane plasmique et que sa spécificité dépendait de la composition de celles-ci. En outre, ces travaux ont permis de révéler un nouveau rôle de la flocculosine dans le pouvoir adaptatif de P. flocculosa, celui de réserve énergétique. Les études de dégradation ont en effet indiqué que le champignon était capable de cataboliser sa toxine à l'aide d'une enzyme dans certaines conditions limitantes. Cela suggère un rôle beaucoup plus étendu que prévu pour la flocculosine; non pas limité à la protection de la niche écologique du champignon mais aussi impliqué au sein de processus énergétiques complexes. Bref, cette étude a démontré le potentiel antimicrobien de la flocculosine tout en jetant un éclairage nouveau sur ses implications biologiques.
