Abstract
L'hémocyanine est la protéine responsable du transport de l'oxygène dans le liquide physiologique (hémolymphe) d'un grand nombre d'invertébrés marins. Chez certaines espèces de mollusques (Megathura crenulata et Concholepas concholepas), l'hémocyanine a démontré d'intéressantes propriétés immunostimulantes. Le projet visait la purification de l'hémocyanine du crabe des neiges (Chionoecetes opilio) par chromatographie d'exclusion moléculaire, sa caractérisation (détermination de la taille moléculaire, du point isoélectrique, la présence ou non d'unités glucosidiques et d'activité phénoloxydase), et l'évaluation de son activité immunostimulante sur des macrophages de souris. Les caractéristiques de l'hémocyanine du crabe des neiges étaient généralement comparables à celles rapportées chez d'autres crustacés. Sur des macrophages de souris, elle a démontré une stimulation de la prolifération et du métabolisme cellulaire, de la production d'oxyde nitrique et de cytokines. Ces effets étaient comparables ou même supérieurs à ceux observés chez les préparations commerciales. Ces travaux permettront de générer des données précises sur l'hémocyanine en vue d'applications biomédicales.
