La physiologie des épithéliums requiert la distribution asymétrique de nombreuses composantes cellulaires. Cette organisation structurale est nommée polarité épithéliale et se caractérise par la formation d'un domaine apical et un domaine basolatéral. La polarisation des cellules épithéliales dépend de la protéine Crumbs 3 (CRB3 chez la souris). Plus spécifiquement, cette protéine contribue à l'établissement du pôle apical et à la formation des jonctions serrées dans les cellules épithéliales en culture. De plus, la protéine CRB3 est capable de réprimer la croissance tumorale et la formation de métastases dans des modèles de xénogreffes. Toutefois, les mécanismes moléculaires agissant en aval de CRB3 sont méconnus et les fonctions physiologiques de CRB3 in vivo chez les mammifères sont peu caractérisées. Ainsi, l'objectif de mon projet était d'analyser le phénotype des souris invalidées pour Crb3 et de valider son rôle potentiel en tant que suppresseur de tumeurs. Nous avons généré des lignées de souris permettant le knockout conditionnel de Crb3. Comme première approche, nous avons invalidé le gène Crb3 dans tout l'organisme. Nous observons une létalité périnatale associée à une détresse respiratoire. Toutefois, les animaux Crb3-/- ne présentent pas de défaut macroscopique majeur. Une analyse plus détaillée révèle des anomalies de développement des alvéoles pulmonaires, la présence de kystes rénaux et la fusion des villosités intestinales. CRB3 est également essentielle pour maintenir l'identité de la membrane apicale dans les cellules épithéliales rénales et intestinales. Une perte de CRB3 cause également une forte augmentation des niveaux de β-caténine cytoplasmique dans l'épithélium intestinal. Cette protéine est un médiateur essentiel de la voie oncogénique Wnt. De plus, la mutation de Crb3 amplifie la croissance tumorale chez les souris ApcMin/+, qui développent de nombreux adénomes dus à la suractivation de la voie Wnt. Dans l'ensemble, nos résultats mettent en lumière l'importance de CRB3 dans l'homéostasie de l'épithélium pulmonaire, rénal et intestinal. De plus, nos données suggèrent que CRB3 pourrait agir en tant que suppresseur de tumeurs via son action sur la voie Wnt.
