Abstract
Tanaman obat Phyllanthus niruri L telah diketahui mempunyai efek sebagai obat anti alergi. Dalam upaya mengoptimalkan penggunaan tanaman tersebut sebagai obat anti alergi dilakukan fraksinasi ekstrak simplisia. Tujuan penelitian ini adalah untuk menilai fraksi mana dari Phyllanthus niruri L herba yang berperan optimal sebagai anti alergi pada mencit dengan dermatitis alergika dan membuktikan adanya senyawa flavonoid dalam fraksi aktif Phyllanthus niruri L herba secara kualitatif. Uji efek anti alergi menggunakan 35 ekor mencit jantan dewasa galur Swiss Webster yang dibagi menjadi 5 kelompok (n=7) yaitu kelompok I (fraksi n-heksan), kelompok II (fraksi etil asetat), kelompok III (kontrol pembanding), kelompok IV (kontrol negatif), dan kelompok V (kontrol positif). Masing-masing kelompok diinduksi ovalbumin pada hari ke 1,7, dan 21, kecuali kelompok kontrol negatif. Bahan uji dengan fraksi n-heksana dan etil asetat diberikan pada hari ke 21 secara oral. Data yang diukur persentase jumlah eosinofil pada apus darah tepi. Data dianalisis menggunakan ANAVA yang dilanjutkan dengan uji beda rata-rata 2-tail p-values for pairwise t-tests dengan α = 0,05 menggunakan program MegaStat v 7.11. Uji kualitatif dilakukan dengan menggunakan metode KLT dengan pelarut CHCl3 : Metanol (9:1), larutan pendeteksi bercak AlCl3 10% dan pelarut Heksan : Etil asetat (1:1), larutan pendeteksi bercak H2SO4 10%. Hasil penelitian uji beda rata-rata dengan 2-tail p-values for pairwise t-tests menunjukkan persentase eosinofil dari kelompok I (4,86) dibandingkan dengan kelompok V (16,57) menunjukkan perbedaan yang sangat signifikan (p=0,0001). Sedangkan kelompok II (12,29) dibandingkan dengan kelompok V (16,57) menunjukkan perbedaan yang non signifikan (p=0,1271). Uji kualitatif fraksi Phyllanthus niruri L herba didapat bercak senyawa uji yang sama dengan senyawa banding kaempferol dan flavone yang merupakan senyawa golongan flavonoid. Kesimpulan dari penelitian ini adalah fraksi Phyllanthus niruri L herba yang mempunyai khasiat sebagai anti alergi adalah fraksi n-heksana dan dalam fraksi tersebut mengandung senyawa aktif golongan flavonoid.
