Abstract
Cancer associated fibroblasts (CAFs) are a major cellular component of the tumour stroma and have been demonstrated to promote tumour invasion. However, most in vitro tumour models used to explore the ability of CAFs to promote invasion lack physiological representation or are difficult to analyze. Here, we use TRACER, a novel paper multilayered tumour model, to explore the contribution of CAFs on the invasion of two head and neck squamous cell carcinoma (HNSCC) cell lines. In this thesis, we demonstrate that multiple patient derived CAF lines promote the invasion of tumour cells in TRACER. Furthermore, we identify remodeling of the extracellular matrix (ECM) as the mechanism by which CAFs promote this invasion for one HNSCC cell line. We have demonstrated that the TRACER platform will be a valuable tool for further exploration of tumour-CAF interactions in the context of tumour invasion.
