P1‐425: ALZHEIMER'S DISEASE AND Aβ EFFECT: ELECTROPHYSIOLOGY ON HIPPOCAMPAL NEURONS

Abstract
Alzheimer's disease (AD), the most common type of dementia, is caused by the accumulation of Amyloid beta (Aβ) plaques and Tau tangles, which impairs synapse and brain physiology. The cause of the accumulation of Aβ remains elusive, leading to the need for a better understanding of the mechanism behind AD to find and fix the cause of this condition. Isolation and culture of hippocampal neurons were performed. Neurons were exposed to amyloid β 1–42 (Aβ1–42) and amyloid β 1–38 (Aβ1–38) treatments for 24 h. After Aβ peptide exposure, patch clamp electrophysiology and immunocitochemistry techniques were assessed to investigate the hippocampal neurons electrophysiology and structure. Aβ (1-42) produces a decrease in outward current of hippocampal neurons, causing a decrease in their electrophysiological activity, and – eventually - neuronal cell death. This decrease in current is time- and dose- dependent. Our research demonstrated that Aβ (1-38) neutralized the Aβ (1-42) toxicity and recover the normal physiology of hippocampal neurons. Also, our results have shown that Aβ effect is not specific for central nervous system cells. Aβ plaques are the key to understand the mechanism that triggers neurodegeneration in AD. Aβ (1-38) inhibits the effect of Aβ (1-42), suggesting a potential treatment to block Aβ effect and recover the electrical activity of central nervous system cells.
