Oxidative Stress Mediates the Trasnformation of Rodent Hepatic Stellate Cells to Myofibroblasts in Culture

Abstract
HSC are heterogeneous regarding the expression of Hedgehog (Hh) and Wnt, pathways activated during development. Also, the Hh pathway is up‐regulated during HSC activation ( Lab Invest 2005, 85:1368–80). We now investigated expression of the Hh and Wnt pathways during HSC activation in culture. Activation of HSC is reversible when cells of early passages are stored in liquid N2, as when re‐plated, mRNA levels of PDGF‐β and α‐SMA are very low but are up‐regulated with a maximum increase by 5 days. This increase is blunted by catalase and N‐acetylcysteine. HSC activation is also accompanied by upregulation of mRNAs for the Wnt pathway members N‐cadherin, β‐catenin, GSK 3β and lymphoid enhancer factor, events that are reproduced by PDGF‐BB. ROS production and expression of α‐SMA mRNA by cultured HSC obtained from Rac‐transgenic mice are elevated as compared to control HSC. Rac HSC have increased Hh activity as evidenced by increased transcription of Patched, and overexpression of Smoothened and Gli2/3. These findings establish a direct connection between ROS and HSC activation. We propose that differentiation and not activation should be used to define the transformation of HSC to myofibroblasts.
