Abstract
La lutte contre la pourriture à sclérotes du soya nécessite le développement de méthodes d'inoculation pour évaluer la résistance des nouvelles lignées à la maladie. Dans le cadre de ce projet, une nouvelle méthode d'inoculation artificielle (méthode du coton imbibé d'une suspension de mycélium), développée antérieurement dans notre laboratoire pour emploi en serres, a été optimisée pour permettre son emploi en conditions de champ. Pour ce faire, deux stades de développement des plantes au moment de l'inoculation et la nécessité ou non de l'irrigation ont été testés. Les résultats ont montré qu'un système d'irrigation n'est pas obligatoire, bien qu'il améliore la capacité de discrimination et contribue à assurer une pression de maladie constante. Quant au stade d'inoculation, les deux stades employés (RI et R3) se sont avérés équivalents. Une fois optimisée, cette méthode a été comparée à la méthode des sclérotes, laquelle est utilisée par le Réseau des grandes cultures du Québec (RGCQ) lors des essais officiels d'enregistrement. La méthode du coton imbibé s'est avérée reproductible et significativement corrélée avec la méthode des sclérotes. Elle a montré une discrimination très fine des cultivars résistants et sensibles dans tous les essais, même lors d'un été chaud et sec, ce que n'a pas permis la méthode des sclérotes.
