Abstract
Epidermal growth factor receptor (EGFR)-activating mutations will be found in 10% to 15% of advanced non–squamous non–small-cell lung carcinoma for white patients and have been associated with a dramatic response to EGFR tyrosine kinase inhibitors (TKIs). All patients treated with first- or second-generation EGFR TKIs will experience disease progression because of secondary mutations (T790M mutation in exon 20, which confers sensitivity to third-generation EGFR TKIs), phenotypic changes, and activation of bypass signaling pathways.
