Effects of donepezil hydrochloride on cognitive function and serum IGF-1 level in patients with vascular dementia

Abstract
Objective To observe the effects of donepezil hydrochloride on cognitive function and serum insulin-like growth factor-1 (IGF-1) level in patients with vascular dementia. Methods A total of 48 patients with vascular dementia admitted to the Binzhou Center Hospital from January 2017 to September 2018 were divided into experimental group (donepezil hydrochloride treatment group) and control group (conventional treatment group) according to the random number table method, with 24 cases in each group. The clinical efficacy, cognitive function scores including mini-mental state examination (MMSE) Montreal cognitive assessment scale (MOCA), daily living ability (Barthel index score), serum IGF-1 level and hemodynamic indexes (plasma viscosity, whole blood high-shear reduced viscosity, whole blood low-shear reduced viscosity, hematocrit, fibrinogen) before treatment and 2 months after treatment were compared between the two groups. Results The scores of MOCA, MMSE and Barthel index in the two groups were higher than those before treatment 2 months after treatment (P all<0.05), and the changes of above indexes were more significant in experimental group (P all<0.05). The total effective rate of experimental group was significantly higher than that of control group (P<0.05). The plasma viscosity, whole blood high-shear reduced viscosity, whole blood low-shear reduced viscosity, hematocrit and fibrinogen level in the two groups were lower than those before treatment, while the IGF-1 level was higher than that before treatment, and the changes were greater in experimental group (P all <0.05). Conclusions Donepezil hydrochloride for vascular dementia can enhance the serum IGF-1 level and improve hemodynamic indexes, thus enhance efficacy and improve cognitive function and daily living ability.
