Introducción: El cáncer es un conjunto de enfermedades en las que células anormales se multiplican e invaden otros tejidos sin control. Uno de los tratamientos más comunes del cáncer es la radioterapia, en la cual se irradia el área cancerosa para destruir las células tumorales y reducir el crecimiento. En la radiobiología, que estudia los fenómenos que ocurren en los seres vivos tras ser sometidos a radiación, se utilizan los ensayos clonogénicos. Para su análisis se requiere del conteo manual de colonias, formadas por células cancerosas que mantienen su capacidad reproductora. Sin embargo, este proceso es tedioso, impreciso, subjetivo y toma mucho tiempo. Objetivo: Desarrollar una nueva técnica basada en el procesamiento digital de imágenes para realizar el conteo automático de colonias celulares y evaluar su exactitud diagnóstica para identificar colonias de células tumorales. Materiales y métodos: Se utilizaron cultivos celulares de las líneas tumorales de seno MCF7 y glioblastoma U87. Se realizaron ensayos clonogénicos exponiendo las células tumorales a radiación; tras 14 días de crecimiento, se fijaron y tiñeron las colonias celulares. Con las imágenes obtenidas por escaneo de los ensayos, utilizando el Software ImageJ y aplicando técnicas de filtrado, umbralización, binarización de colores, transformación y etiquetado se desarrolló el método de conteo automático; se añadió una interfaz para permitir la edición manual del conteo. La valoración de la exactitud diagnóstica del método desarrollado se realizó definiendo como estándar de referencia el conteo manual de colonias. Resultados: El método desarrollado mostró un buen rendimiento diagnóstico para la detección de colonias de células tumorales con una sensibilidad del 98,62% (IC 95%: 98.30% - 98.99%) y una especificidad de 81,07% (IC 95%: 78.93% - 83.21%). La detección de colonias tumorales tuvo una exactitud diagnóstica del 95,7% % (IC 95%: 95.26% - 96.17%). Discusión y conclusiones: Se propone el conteo automático de colonias celulares como una herramienta útil en las investigaciones biomédicas sobre el cáncer. El método desarrollado demostró una adecuada eficiencia diagnóstica. Estudios posteriores podrán indagar la precisión y reproducibilidad del método propuesto.
