The role of innate lymphoid cells (ILCs) in a murine model of irritant-induced asthma

Abstract
<b>Rationale:</b> Innate lymphoid cells (ILCs), part of the innate immune system, produce IFN-γ (ILC1), IL-5 and IL-13 (ILC2), and IL-17 (ILC3). The purpose of the present study was to examine the function of ILCs in irritant-induced airway inflammation and function. <b>Hypothesis:</b> We hypothesized that ILCs have a protective role in irritant-induced airway dysfunction and inflammation. <b>Methods:</b> We quantified lung ILC subtypes (ILC; CD45+ Lineage- CD90+ CD127+) and T-bet+ ILC1, GATA3+ ILC2 and RORγt+ ILC3 by flow cytometry at 3, 12, and 24 h after chlorine (Cl2) exposure in BALB/c mice. We assessed airway responsiveness to methacholine (FlexiVent) in RAG-/- mice, deficient in T and B cells, after depletion of ILCs with anti-CD90 antibody (i.p) 12h before and 3h after Cl2 exposure and in combined RAG and IL-2rg knockout (RAG-/-IL2rg-/-) mice, deficient in T, B and ILCs. We expanded the lung ILCs with intranasal recombinant IL-33 for 3 successive days in RAG-/- mice and 1 day later we exposed mice to Cl2 and assessed lung function at 24 hours after exposure. In these mice we neutralized IL-13 with an anti-IL-13 antibody i.p at 1h before Cl2 exposure following IL-33-induced ILC expansion. <b>Results:</b> ILC2s and ILC3s were significantly increased 3 and 24h after Cl2 exposure. Lack of ILCs did not affect airway hyperresponsiveness (AHR) and airway inflammation in the response to Cl2. IL-33 expanded ILC2, attenuated AHR and neutrophilia but increased eosinophilic inflammation. IL-13 neutralization restored AHR to methacholine of the Cl2 exposed control group. <b>Conclusions:</b> ILCs and the IL-33/IL-13 pathway may have a protective role in the prevention of AHR and airway neutrophilia following Cl2.
