Abstract

酒米の重要な特性である蒸米の酵素消化性は,澱粉の糊化温度が低い米において優れる.著者らは,代表的な酒造好適米を含む41品種・系統から,澱粉の糊化温度の指標となる粘度上昇開始温度が低い「秋田酒44号」を見出した.この系統のアミロペクチンの側鎖長分布を解析したところ,「あきたこまち」や「五百万石」等と比べ,重合度6から12の短鎖の比率が高く,重合度13から24の中鎖の比率が低かった.また,同系統は胚乳における澱粉代謝系酵素のひとつ,澱粉ホスホリラーゼ1の活性を欠損していた.一方,「秋田酒44号」とその系譜上にある品種・系統について,澱粉ホスホリラーゼ1の活性を調査したところ,活性の欠損は「秋田酒44号」以外には認められなかった.「秋田酒44号」は「58系3071」へのγ線照射によって育成されていることから,澱粉ホスホリラーゼ1活性欠損の原因は,このγ線照射による変異が原因と推定された.さらに,「秋田酒44号」の澱粉ホスホリラーゼ1遺伝子の塩基配列を決定し,同酵素活性を保持する「日本晴」の配列と比較したところ,第12エクソンにアミノ酸置換を伴う一塩基置換があることを確認した.この変異が「秋田酒44号」の澱粉ホスホリラーゼ1活性の欠損,ひいては澱粉糊化時の粘度上昇開始温度が低い原因であると考えられた.
