Abstract

The deoxyribonucleic acid (DNA) is a convenient molecule for the rational design of molecular switches and is ideally suitable for the development of molecular computing systems. Throughout this chapter, we discuss how one can rationally design DNA switches to engineer logic circuits. This linear process starts from the selection of a specific molecular input and its related DNA-based recognition element that is further engineered into a structure-switching nanosystem. Then, an output function is introduced and optimized to enable analytical readout of the switching response. Finally, we provide several rational strategies for the implementation of these DNA switches into logic circuits and discuss upcoming challenges regarding real-life applications.
