Hypertension and CaMKII isoforms in the endothelium of murine mesenteric arteries

Abstract
Crucial to endothelial functions, Ca 2+ homeostasis in native endothelial cells has been studied using hypertensive mice (chronic infusion of phenylephrine; PE). Localized spontaneous endothelial Ca 2+ oscillations, termed Ca 2+ pulsars, appear to be altered in hypertensive mice. Interestingly, calmodulin‐dependent protein kinase II (CaMKII) is involved in endothelial function and is uniquely characterized by its ability to decode oscillatory Ca 2+ signals such as Ca 2+ pulsars. The objective of this study is to determine the impact of Ca 2+ pulsars stimulation in hypertension on CaMKII isoforms in endothelial cells from mesenteric arteries. The expression of all four CaMKII isoforms (α, β, δ and γ) is significantly decreased (47–57%) in hypertensive mice compared to controls. Western Blot revealed the presence of CaMKIIα, β, δ but not γ in mesenteric arteries. Consistently, CaMKIIα, β and δ were specifically detected in endothelial cells by immunofluorescence experiments. Furthermore, hypertension is correlated with an exacerbated distribution of endothelial CaMKII in clusters compared to control. Additionally, confocal imaging showed that acute stimulation of Ca 2+ pulsars with PE stimulates clustering of CaMKII in both groups. Our data suggest that endothelial CaMKII can play a significant role in hypertension. Supported by CIHR, FRSQ, CFI and HSFC.
