Loss of murine TRPV1 function alters the thermoregulatory and ventilatory response to acute hypoxia

Abstract
Hypoxic hypometabolism is the process in which small mammals and neonates reduce metabolic rate and body temperature in response to hypoxia. Since the transient receptor potential vanilloid 1 (TRPV1) ion channel has been implicated in the regulation of body temperature, we hypothesized that loss of TRPV1 function would affect the thermoregulatory and ventilatory response to hypoxia. Mutant TRPV1 −/− (KO) and wild‐type TRPV1 +/+ (WT) mice were exposed to hypoxia (10% oxygen) for 4 hours. Body temperature (Tb), heart rate (HR) and activity (ACT) were recorded using telemetry in conjunction with pattern of breathing measurements using plethysmography. During hypoxia, KO mice displayed a significantly lower mean Tb compared to WT mice (31.20 ± 0.13 °C vs. 31.97 ± 0.27 °C; Mann‐Whitney p=0.040). There were also differences in ventilation (1128 ± 36.4 ml/kg/min vs. 1282 ± 59.2 ml/kg/min; 2‐way ANOVA, p=<0.001), and respiratory frequency (161 ± 2 bpm vs. 179 ± 7 bpm; 2‐way ANOVA, p=<0.001) during hypoxia, as well as inspiratory (0.139 ± 0.002 s vs. 0.125 ± 0.003 s; 2‐way ANOVA, p=<0.001) and expiratory time (0.271 ± 0.008 s vs. 0.242 ± 0.015 s; 2‐way ANOVA, p=<0.001) between KO and WT mice, respectively. These data indicate that TRPV1 is involved in the ventilatory and thermoregulatory response to hypoxia. (Supported by CIHR)
