A Role for CD34 in the Response of Skeletal Muscle to Hypoxic Conditions

Abstract
CD34, a satellite cell (SC) marker participates in skeletal muscle (SM) regeneration, facilitating the migration of SC. A fraction of patients with chronic obstructive pulmonary disease (COPD) develop SM atrophy, in part due to increased SC senescence. COPD patients experience transient hypoxemia, and it was suggested these hypoxic episodes may participate in SM wasting. Recently, we have observed that in vitro exposure of SC to hypoxic conditions increased CD34 expression and retained SC in an undifferentiated state. The goal of the present study was to evaluate the importance of CD34 expression in the muscle response to hypoxic conditions in vivo . Total weight and lean body mass were measured in WT and Cd34 knockout ( Cd34 ‐/‐ ) mice exposed to hypoxic conditions (10% O 2 ). The gastrocnemius expression of CD34 and genes involved in muscle regeneration were detected by qRT‐PCR. Results show an important loss of total weight and lean body mass at 2 days, which is significantly worse in Cd34 ‐/‐ mice. Alterations in Mrf4 and MHC expression were observed in Cd34 ‐/‐ mice vs. WT, suggesting a role for this protein in the response of muscle to hypoxic conditions. All in all, our results suggest that CD34 expression is important for the response of SM to hypoxic conditions, leading to a possible role in SM wasting in COPD.
