NPM new interacting partners and influence of NPM mutations on gene regulation

Abstract
Nucleophosmin (NPM) is a multifunctional protein. NPM mutation is found in one third of all acute myeloid leukemia (AML), and in more than 60% of AML with normal karyotype. It is also a recurrent translocation partner in AML. We got interested in the NPM function as a co‐regulator of gene expression. We hypothesized that the modifications of NPM disrupt gene regulation and thereby, favour leukemia. With a combination of biochemistry and molecular biology techniques, we identified new partners of NPM involved in gene regulation. We found that NPM colocalise with different transcription regulators at specific genes. Based on our results, we propose that the effect of NPM modifications on gene regulation has a significant impact on the capacity of hematopoietic cells to receive extra‐cellular signals which favours abnormal hematopoiesis and potentially, leukemia.
