Abstract

Phosphoryl transfer reactions are central to the energy balance of all organisms in modern biology. The chemical potential energy stored inside phosphate bonds is used to drive thermodynamically unfavorable processes crucial for basic metabolic pathways and cellular signaling. Through in vitro selection, a set of phosphoryl transfer ribozymes have been isolated that provide insights into how RNA rather than proteins might have catalyzed phosphoryl transfer in a hypothetical RNA world. In this chapter, we discuss the different mechanisms by which ribozyme phosphoryl transfer reactions occur and compare them with their protein equivalents in modern biology. By exploring kinase, glycosidic bond forming, capping, ligase, and polymerase ribozymes, we highlight the importance of phosphoryl transfer reactions from nucleotide and nucleoside metabolism to the assembly and replication of RNA molecules in an RNA world setting.
