Blockade of immune‐cell vasopressin receptors by V1a antagonist inhibit the peak of the experimental autoimmune encephalomyelitis (EAE) in Lewis rats.

Previous experiments established that in Lewis rats surgical ablation of the neurointermediate pituitary lobectomy (NIL) significantly decreases the clinical and central nervous system histopathological signs of EAE, whereas desmopressin (DP), a synthetic analog of vasopressin (VP) administration to NIL animals, restores the susceptibility to EAE. The presence of vasopressin receptors and AV stimulatory actions on immune cells has been described. If VP from the neural lobe is involved in the EAE response, the blockade of the VP receptors in immunocytes with a VP V1a antagonist (Cat V1880, Sigma), will protect the normal rats against the EAE. Two groups of 8 female rats were used. I.M. injections of saline solution or antagonist AV receptors (0.75 μg/Kg B.W. each 12 hours) started one week before the EAE immunization. Results showed that antagonist treatment induced a significant decrease in EAE clinical score during the peak of the disease (2.2±0.7 vs. 0.8±0.2 p<0.05). The degree of protection against EAE is similar to that observed in NIL rats. Our findings strongly support the concept that VP plays an important role in the EAE physiopathology.
