Grün and Schwalbe review current experimental approaches to study the folding processes of DNA and RNA G-quadruplexes.Folding of nucleic acids with non-canonical secondary structures is an intriguing yet enigmatic process.In particular, G-quadruplex-forming DNA sequences tend to fold into several distinct conformations.This structural polymorphism gives rise to complex folding and refolding dynamics.In the Review the authors discuss feasible folding mechanisms and describe possible folding pathways along the conformational energy landscapes.The consequences of these recent biophysical insights are evaluated using examples of biologically relevant regulatory G-quadruplex elements in oncogene promoters.
