Abstract
The demonstration of increased levels of inflammatory and immune system proteins in Alzheimer’s disease (AD) brain tissue [1-3], combined with epidemiological evidence that anti-inflammatory therapy may decrease the risk of AD [4], has led to the hypothesis that inflammation plays an important role in the disease pathogenesis. As the resident immune-competent cells in the central nervous system (CNS), microglia are the most likely candidate to orchestrate this inflammatory response, by producing and responding to a variety of immune system molecules [2 5 6]. As phagocytes, microglia may also be involved in either the formation or degradation of amyloid, a process important in the physical transformation of senile plaques (SP) [7]. Finally, microglia are a potential source of several neurotoxic substances which could contribute to neurodegeneration [2 8]. Given that microglial cells may play such a central role in AD, it seems prudent to consider therapeutic strategies directed towards manipulating microglial activity in the treatment of AD.
