Fgl2, has been implicated in the pathogenesis of viral-induced hepatitis, cytokineinduced fetal loss syndrome, allograft and xenograft rejection.This 432-amino-acid protein, a member of the fibrinogen-related protein superfamily was originally cloned from a mouse cytotoxic T lymphocyte cell line.Preliminary findings from our laboratory suggest that fgl2 exists both as a transmembrane protein and a secreted form with a truncated N-terminus.Studies demonstrate that fgl2 expressed on the membranes of murine hepatitis type-3 infected macrophages and endothelial cells exhibit prothrombinase activity.In contrast, no coagulant activity was detected from the T cell secreted form of fgl2.Since proteins belonging to the fibrinogen-related protein superfamily are known to exert multifaceted roles in the immune and inflammatory response, we postulated that secreted fgl2 exerts immunomodulatory activity.To examine this, we overexpressed fgl2 protein in a baculovirus expression system and studied secreted protein for its effect on primary immune responses.Purified fgl2 protein inhibited T lymphocyte proliferation in allogeneic, anti-CD3 antibody, and Con A-stimulated murine spleen cells in a dose-dependent fashion.This was inhibited by monoclonal antibodies against mouse fgl2.Decreased levels of IL-2 and IFNgamma and increased levels of IL-4 and IL-10 were observed in supernatants from an allogeneic mixed lymphocyte reaction to which purified fgl2 protein had been added.Furthermore, fgl2 protein prevented LPS-induced maturation of bone marrow-derived DC by decreasing the expression of the co-stimulatory molecules CD80 and CD86.These results provide the first evidence to demonstrate that fgl2 exerts anti-inflammatory properties with an inhibitory effect on T lymphocyte activation via promotion of a Th2 cytokine profile and inhibition of dendritic cell maturation.
