Abstract
This chapter focuses on recent developments of both synthetic and natural photoswitches intended for use in vivo. Synthetic photoswitches fall into several groups based on the type of photochemistry they undergo. In general, to be suitable tools for manipulating cells, photoswitches must be stable in cellular environment and undergo photochemistry at wavelengths compatible with cells and tissues at moderate light intensities. Several photoswitches undergo photochemistry at short ultraviolet wavelengths, a feature that is less critical for those interested in optical memory storage for example. Applying chemically modified biomolecules in vivo requires the photoswitch be delivered to appropriate sites. Perhaps the best-known natural light-sensitive protein is rhodopsin, a protein found in the retina. The development of natural photoswitches as tools seems likely to grow quickly as new light sensitive modules are discovered via genome sequencing efforts.
