Adrenalectomy Does Not Prevent Exercise-Induced Intestinal Lymphocyte Depletion Following Treadmill Exercise In Mice

Abstract
0909 Lymphocytopenia is a common occurrence following strenuous exercise that may be responsible for the post-exercise immunosuppression observed in athletes and exercise participants. This increased lymphocyte loss may be due to increased apoptosis mediated by adrenal hormones, especially glucocorticoids (GC), produced during strenuous exercise. Purpose: To examine the role of adrenal hormones in intestinal lymphocyte (IL) loss following strenuous exercise in mice. Methods: Thirty-six female C57BL/6 mice were randomly assigned to receive bilateral adrenalectomy (ADX) or control (SHAM) surgery. One week following surgery mice were randomly assigned to (i) a control condition (nonexercised) in which mice were exposed to treadmill noise and vibration but not running, (ii) treadmill exercise (90min, 28m·min-1, 2° slope) and sacrificed immediately following exercise (immediate), or (iii) treadmill running and sacrificed 24h following running (24h Post). ILs were isolated and T and natural kill (NK) subpoulations were determined by flow cytometry. Plasma corticosterone (CORT) was determined by RIA. Results: Plasma CORT was virtually nil in ADX treated mice (mean ± SEM; 3.1 ± 0.6ng·ml−1). Total IL, CD3αβ+, CD3γδ+, CD8αα+, and NK+ cells recovered from the bowel were significantly higher in ADX (28.4 ± 2.8 × 106; 11.2 ± 1.1 × 106; 10.4 ± 1.6 × 106; 19.7 ± 2.4 × 106; 1.7 ± 0.3 × 106 cells) compared to SHAM mice (21.2 ± 2.2 × 106; 8.5 ± 0.8 × 106; 5.9 ± 0.7 × 106; 11.7 ± 1.1 × 106; 0.9 ± 0.2 × 106 cells; p < 0.05 in each case). Although ADX produced some sparing effect across exercise condition, ADX did not significantly inhibit exercise-induced cell loss. 24h after treadmill exercise there were significantly fewer ILs (16.3 ± 1.8 × 106), CD3αβ+ (7.2 ± 0.8 × 106), and CD8αα+ (8.2 ± 1.2 × 106) cells compared to nonexercised mice (28.4 ± 3.1 × 106; 11.1 ± 1.3 × 106; 18.8 ± 2.6 × 106, respectively; p < 0.05 in each case). Conclusion: Although ADX buffered some IL loss relative to SHAM, the absence of adrenal hormones (most notably GC) did not reverse the exercise-induced intestinal lymphocyte loss observed at 24h post exercise. These data suggest IL depletion observed after strenuous exercise in mice is not mediated by adrenal hormones, at least not directly, and that other mechanisms are likely involved.
