Abstract
RAC1 is a GTPase that plays an evolutionarily conserved role as a “molecular switch” coordinating cellular innate immune defenses from plants to mammals. Chronic inflammatory bowel diseases are associated with dysregulation of innate immune defenses. Here we report a novel genetic association of RAC1 with IBD. An initial candidate gene approach of RAC1 was used with 12 tag SNPs in a discovery cohort consisting of 2281 Caucasian subjects including 754 CD and 603 UC patients, and 924 controls. Results were confirmed in a first cohort comprised of 1836 Caucasian subjects including 443 CD and 477 UC patients, and 916 controls recruited from North America and a second cohort from Scotland comprised of 2449 Caucasian subjects including 691 CD and 615 UC patients, and 1143 controls. Functional studies were carried out using Rac1 conditional phagocyte knockout mice. Rac1-KO and littermate control mice were treated with dextran sodium sulphate (DSS).
