Abstract
Rationale: Oxidative stress associated with asthma pathobiology targets phospholipids in lung cell membranes, and airway surfactant fluid.We have shown that a panel of oxidized phosphatidylcholines (OxPC) accumulates in the lungs of mice and humans after inhaled allergen challenge, and associates with the severity of airway hyperresponsiveness.This is, in part, due to direct effects on airway smooth muscle, as OxPC triggers an acute increase in intracellular Ca 2+ [Ca 2+ ] i in cultured human airway smooth muscle cells (HASM).Here, we test the hypothesis that OxPC directly induces airway narrowing, and correlates with the activation of a unique repertoire of pathways that regulate intracellular Ca 2+ flux in HASM.Methods: We used murine thin cut lung slices (TCLS) and phase contrast video microscopy to assess airway narrowing (n=3 mice).Changes in [Ca 2+ ] i in HASM cells
