Abstract
vitro translation system supplemented with semipermea- bilized cells (Wilson et al. 1995). 35S-labelled translation products were treated with MMP-2, MMP-9, MMP-12, MMP-13 and MMP-14.The sixth TB module and flanking domains of fibrillin-1 (3fib-1), profib-1 and glyfib-2 were generated in a mammalian expression system using COS-1 cells and purified recombinant proteins treated with MMP-2, MMP-9, MMP-12 and MMP-13.Digestion products were characterized by N-terminal automated amino acid sequencing. ResultsnFib-1 and mFib were cleaved by MMP-2, -9, -12 and -13 generating a major cleavage product of ~45 kD.Cleavage of profib-1 and glyfib-2 generated a product of ~12 kD.Amino acid sequencing of profib-1 and glyfib-2 MMP cleavage fragments showed the cleavage site to be at exon junction 10/11 region.cFib-1 was cleaved by each of the MMPs with removal of ~20 kD and scFib-1 to a single major product of ~60 kD.MMP-12 treatment of 3fib-1 localized the cleavage site to the region of the sixth TB domain.Amino terminal sequencing showed the MMP cleavage site to be at the carboxy-terminal end of exon 49. DiscussionUsing a recombinant approach, we have identified major MMP degradation products of nFib-1, mFib-1 and scFib-1.We have defined three cleavage sites at the exon 10/ 11 junction of fibrillin-1 and 2, and the carboxy-terminal end of exon 49 in fibrillin-1.MMP cleavage of microfibrils may be crucial in the physiological and pathological turnover of microfibrils in connective tissues.
