Identifizierung einer prädiktiven Markersignatur zur Diagnose von HNSCC basierend auf Thrombozyten-RNAseq

Abstract
Einleitung Die ‚liquid biopsy‘ bietet eine Möglichkeit, Krebs durch Untersuchung von Körperflüssigkeiten zu identifizieren. Unsere Studie beschäftigt sich mit der Analyse von ‚tumor-educated‘ Thrombozyten, einer kürzlich entdeckten neuen Option der ‚liquid biopsy‘. Frühere Forschungen identifizierten eine Tumor-Thrombozyten-Interaktion, die zu einem Transfer von tumorabgeleiteter RNA in Thrombozyten führte, die deshalb auch als ‚tumor-educated platelets‘ (TEP) bezeichnet werden. Material und Methoden RNA aus Thrombozyten von Tumorpatienten und gesunden Spendern (n=5/5) wurde extrahiert und mittels RNAseq analysiert. Zusätzlich wurde RNA aus dem entsprechenden Tumor sequenziert. Bioinformatischer Tools wurden angewendet. Die unterschiedlich vorhandene mRNA in Thrombozyten von Tumorpatienten im Vergleich zu gesunden Spendern wurde in einer zweiten Kohorte (n=6/7) mittels quantitativer RT-PCR verifiziert. Ergebnisse RNAseq-Daten zeigten 426 signifikant unterschiedlich existierende RNA in Thrombozyten gesunder Spender versus Thrombozyten von Tumor-Patienten. Unter ihnen identifizierten wir 49 RNA, die für charakteristische Epithelzellen-Gene kodieren. Darüber hinaus beobachteten wir in Thrombozyten von Tumorpatienten RNA, die für Gene kodieren, die an der Tumorprogression beteiligt sind, indem sie zur Proliferation, Metastasierung oder Angiogenese beitragen. Wir identifizierten 5 differenziell vorhandene mRNA als potenzielle ‚liquid biopsy‘ Biomarker in TEP. Schlussfolgerung Basierend auf diesen vielversprechenden Ergebnissen dieser Pilotstudie sollte eine prospektive Studie mit einer größeren Kohorte initiiert werden, um die hier vorgeschlagene prädiktive Markersignatur zu verifizieren, die eine Identifizierung von HNSCC anhand von Thrombozyten-RNAseq ermöglicht.
