Optimized seed bank management requires a representative\nportrait of the true quality of the conserved seedlots. This\nimplies knowing how long germination test results remain\nvalid. To validate the empirical grid for germination test\nfrequency which was based on seedlot history at the Berthier\nTree Seed Centre (Québec), seed sources were identified\nand harvested repeatedly; seedlot germination was then\nassessed annually. The principal coniferous species used\nin Quebec for reforestation were examined: white spruce\n(Picea glauca [Moench] Voss), black spruce (P. mariana\n[Mill.] B.S.P.), Norway spruce (P. abies [L.] H. Karst), white\npine (Pinus strobus L.). and jack pine (P. banksiana Lamb.).\nSeed water status is a key element for their conservation.\nWater activity measurement characterizes seed water status\nand its possible evolution more precisely than water content\nmeasurement. This new tool has three main advantages: it\nis fast, easy to obtain and, above all, non-destructive. Water\nactivity measurements revealed a significant change in the\nwater status of the studied seedlots, as a consequence of\nthe repeated openings of the containers. Though it did not\nresult in a reduction in germination, this change in seed\nwater status altered the seedlot conservation conditions.\nThe authors discuss the increasing role of water activity\nin seed bank management and the precautions required\nwhen sampling in conservation containers. / La gestion optimisée d'une banque de semences requiert un portrait représentatif de la qualité réelle des lots conservés. Il importe d'estimer la durée de validité d'un test de germination. Pour valider la grille empirique de fréquence des tests basée sur l'historique des lots entreposés au Centre de Semences Forestières de Berthier (Québec), des sources de semences ont été identifiées et récoltées à ,plusieurs reprises, puis leur germination a été évaluée annuellement. Les principales essences de reboisement au Québec ont été visées : l'épinette blanche (Picea glauca [Moench] Voss), l'épinette noire (P. mariana [Mill.] B.S.P.), l'épinette de Norvège (P. abies [L.] H. Karst.), le pin blanc (Pinus strobus L.) et le pin gris (P. banksiana Lam.).\nL'état hydrique des semences est un élément clé pour leur conservation. La mesure de l'activité de l'eau (AE) permet de caractériser, plus finement que la teneur en eau, l'état hydrique des semences et son évolution. Ce nouvel outil a l'avantage d'être rapide, facile et surtout, non destructif. Des mesures d'AE effectuées sur les lots du projet sur la fréquence des tests ont révélé une modification significative de leur état hydrique, conséquence des ouvertures répétées des contenants. Bien qu'elle n'est influencé la germination, cette modification de l'état hydrique a altéré les conditions de conservation des lots. Les auteurs discutent du rôle grandissant de la mesure de l' AE dans la gestion des banques de semences et des précautions à prendre lors des prélèvements dans les contenants en conservation.
