The diagnostic potential of genome-wide sequencing is limited by our ability to interpret rare and novel variants. Consequently, germline missense variants are often classified using current guidelines as having uncertain clinical significance. New scalable and easy-to-implement strategies are needed to generate supporting evidence for pathogenicity. Somatic cancer driver mutations that have functional consequences are sometimes observed as germline missense variants. We hypothesized that publicly available cancer mutational data can be used to develop and test a new statistical method of predicting the pathogenicity of germline missense variants.
