Abstract This chapter describes the procedure for synthesis of ethoxypillar[6]arene. It presents some of the important points to be considered, the conditions that need to be maintained, characterization data, and the reagents required, as well as the techniques used and the equipment setup that are vital to carrying out the process. The chapter also describes the hazards associated with working with chemicals and the ways to deal with these hazards. Pillar[n]arenes are a family of macrocyclic hosts first reported in 2008 that have quickly become an important scaffold in supramolecular chemistry. First made in 2009, pillar[6]arene has become a highly desired analog due to its ability to bind larger, biologically relevant guest molecules.
