INTERAÇÃO ENTRE IL-1Α E TGF-Β1 NA CICATRIZAÇÃO NORMAL E HIPERTRÓFICA

Abstract
Resumo: A cicatrização é um processo complexo, estritamente regulado celular e molecularmente. A menor perturbação desse processo pode levar à formação de cicatrizes hipertróficas (Hscars), uma condição patológica de natureza fibrótica. Hscars são caracterizadas por sua aparência avermelhada devido à formação/manutenção de uma extensa rede de vasos sanguíneos (neoangiogênese). No entanto, a característica mais marcante das Hscars é a deposição excessiva de colágeno, sustentada por miofibroblastos que permanecem na área apesar da cicatrização da superfície lesada. Muitas biomoléculas estão envolvidas nesse processo, à exemplo do TGF-β1 e da IL-1α, moléculas com atividade pró-fibrótica e inflamatória, respectivamente. Uma vez estabelecidas, as Hscars podem apenas cicatrizar parcialmente ao longo do tempo, resultando na formação de cordões fibrosos ou contraturas que podem comprometer a função do órgão em sua totalidade, ou apenas parcialmente. Até o momento, não existe um tratamento eficaz contra a formação de Hscars. As ferramentas terapêuticas disponíveis restringem-se ao uso de pomadas, tratamentos à laser, ou o uso de roupas compressivas. Em casos extremos de contraturas incapacitantes, a excisão cirúrgica é a abordagem recomendada. Tratamentos não invasivos (pomadas, compressão local etc.) ocasionam o aumento na expressão de IL-1α no sítio da lesão. O objetivo deste trabalho de revisão é caracterizar a ação individual de cada citocina e suas ações concomitantes quando atuam no mesmo espaço temporal, em um contexto de cicatrização.
