Abstract
Obesity is now known to be a disorder of the brain implying dysfunction of the neuronal circuits managing satiety and energy expenditure. The arcuate nucleus of the hypothalamic hosts two neuronal populations highly relevant for energy homeostasis. In a classical model, Agouti-related protein (AgRP) neurons drive appetite and energy sparing. In contrast, pro-opiomelanocortin (Pomc) neurons drive satiety and energy expenditure. Pomc neurons become dysfunctional in a state of prolonged obesity; indeed, roughly 25-50% of them disappear under conditions of long-term metabolic stress.
