Abstract
An automated process optimization platform consisting of synthesis, sampling, and analysis modules for the optimization of multiple process quality attributes is reported. The synthesis module is capable of conducting flow chemistry experiments using microliter-size reaction plugs, which are sent to online analytics equipped with a recirculatory multidimensional chromatographic technology for the measurements of two independent attributes. The onboard analytics is also capable of validating the measurements using a unique recirculatory flow-path in a multidimensional chromatographic workflow. The robotic platform is capable of measuring the yield and stereochemical purity of a target product from a small amount of raw material (specifically, 1.5 mg per experiment) with minimal human assistance.
