Abstract
While the majority of Retinoblastoma (RB) is caused by biallelic inactivation of the RB1 gene, approximately 1.5% of RB occurs as a result of MYCN gene amplification. Unlike RB1 pathogenic variants, which are present in constitutional cells in 100% of bilateral RB patients and 19% of unilateral RB patients, MYCN gene amplification is a purely somatic event and is not associated with hereditary RB. Confirmation that the cause for RB in a proband is due to somatic MYCN amplification eliminates the risk for secondary ocular tumors and life-long hereditary RB-associated secondary cancers for the proband.
