Abstract

The cblG inborn error of vitamin B12 metabolism is associated with pathogenic variants in the MTR gene, which encodes methionine synthase. Over 50 patients with the disorder have been identified, and 53 potentially causal MTR variants have been published. Over several decades we have established the diagnosis in many patients of cblG by somatic cell complementation. With the discovery of the genes involved in the inborn errors of vitamin B12 metabolism most diagnosis is now performed by molecular analysis and complementation analysis is rarely performed.
