Dyskerin pseudouridine synthase 1 (DKC1, OMIM 300126) is known to be associated with X-linked dyskeratosis congenita (DKCX; OMIM 305000) – a bone marrow failure syndrome clinically characterized by reticular skin pigmentation, abnormal nails, and oral mucosal leukoplakia. In addition to that, mutations in the DKC1 gene have been associated with a high risk of developing aplastic anemia, myelodysplastic syndrome, leukemia, and solid tumors. The DKC1 missense mutation, c.5C>T (exon 1), leading to the p.Ala2Val has been reported in a family affected with dyskeratosis congenita.
