Abstract
Mucopolysaccharidosis type 1, or Hurler syndrome (MPS1-H), is a rare lysosomal storage disorder caused by alpha-L-iduronidase enzyme deficiency due to biallelic pathogenic variants in the IDUA gene. MPS1-H is characterized by coarse facies, corneal clouding, intellectual disability, dysostosis multiplex, and hepatosplenomegaly, among other features. This disorder is also part of a phenotypic spectrum, with MPS1-S (Sheie syndrome) representing the milder/attenuated end of the continuum. Bone marrow transplantation and/or enzyme replacement therapy have been shown to prevent or delay some of the features associated with MPS1-H, making it a suitable target for population-level screening.
