Abstract
Este protocolo describe una actividad educativa que tiene como fin explicar las bases de la purificación de proteína con materiales que pueden ser usados sin la infraestructura tradicional de un laboratorio. En pocas palabras, una proteína de interés (en este caso, 6xHis-sfGFP) se produce en extractos cell-free toda la noche. Al día siguiente, se purifica usando el kit NEBExpress Spin Columns y de acuerdo a las instrucciones del fabricante. Todas las etapas de centrifugación (unión, lavado y elución) se realizan usando una centrífuga de mano fabricada con impresión 3D. La actividad dura dos horas. This protocol describes an educational activity to explain the basics of protein purification with materials that can be used without traditional lab infrastructure. In few words, the protein of interest (in this case, 6xHis-sfGFP) is produced in cell-free lysates overnight. The day after, it is purified using NEBExpress Spin Columns and all the centrifugation steps (for binding, washing and elution) are performed using a 3D-printed hand-powered centrifuge. The activity lasts two hours.
