Janus kinase inhibition in cutaneous histiocytosis: A systematic review

Abstract
C utaneous histiocytoses are a group of rare diseases characterized by aggregation of histiocyte cells in the skin and other organs. 1 Although some cases are self-limited, nonresolving events are treated with steroids or chemotherapy.Janus kinase inhibition (JAKi) has recently shown effectiveness in treating refractory cutaneous histiocytoses.This review examines the outcomes of JAKi in cutaneous histiocytosis.Following registration with the International Prospective Register of Systematic Reviews (PROSPERO) (CRD42024573234), a Cochrane, Embase, and MEDLINE search was conducted under the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines using keyword variations of histiocytosis and specific syndromes and JAK inhibitors with specific drug names (Supplementary File 1, available via Mendeley at https://data.mendeley.com/datasets/ybpfytht2p/1).A screening of 588 articles yielded 34 total studies (publication date: 2019-2024) and 51 participants (Supplementary Files 1-3, available via Mendeley at https://data.mendeley.com/datasets/ybpfytht2p/1).The mean age of participants was 29.2 years (range, 0.02-67 years), including 22 males (43.1%) and 18 females (35.3%), with 39 documented JAKi treatment outcomes.Treatments were divided into 2 categories: concomitant medications (66.7%, 26/39), most commonly with methylprednisolone (38.5%, 10/26), or no concomitant medications (3/13, 33.3%).The mean treatment duration was 6.5 weeks (range, 1-39 weeks).Cutaneous histiocytosis disease specified subtypes were reported in 38.4% (15/39) of cases and
