Advanced Upconversion Nanoparticles for Improved Endothelial Barrier Transport

Abstract
Upconversion nanoparticles (UCNPs) have been used in bio-imaging because they can emit light under the excitation with longer wavelength. However, the poor transportation of UCNPs crossing tissue barriers results in inefficient delivery of UCNPs to the targeted organ/tissues. Herein, the core-shell UCNPs modified with cationic biopolymer, i.e., N, N-trimethyl chitosan (TMC), have been developed for improved endothelial barrier transport. The core-shell UCNPs with the average particle size of 26.05±3.72 nm have the emission at 443 nm under the excitation of 908 nm. The core-shell UCNPs have been further modified with biocompatible polymers to enhance water dispersibility, and surface charge positivity. In addition, the transport ability of the core-shell UCNPs has been investigated by using an in vitro endothelial barrier model. The results indicate that TMC modified on the surface of core-shell UCNPs can significantly improve the endothelial barrier transport by two times as compared to core-shell UCNPs without TMC.
