Birth defects are due to a combination of environmental and genetic factors. This chapter provides examples of some common craniofacial, orofacial, and dental anomalies and special treatment considerations required by orthodontists. A patient may present with a confirmed genetic diagnosis, but observant clinical examination is often the initial clue to an important genetic contribution. Consequences of the presence of any single etiologic factor differ from one embryologic environment to another. The complexity of determining contributions of multiple genetic, environmental, and growth interactions to the complex inheritance traits of a Class II or III malocclusion is a challenge. The number of single gene mutations associated with craniofacial anomalies is extensive. Alterations of tooth number, size, and shape are common craniofacial anomalies, while structure and eruption problems are less frequent. Anomalies of tooth structure involve three specialized calcified hard tissues: enamel, dentin, and cementum.
