Abstract

Hintergrund: Hereditäre Neuromuskuläre Erkrankungen (NME) zählen zu den seltenen Erkrankungen und führen oft zu erheblichen Einschränkungen oder vorzeitigem Versterben. Die eingeschränkten Therapieoptionen verdeutlichen die Dringlichkeit innovativer Ansätze zur Diagnostik und Therapie. Das EU-geförderte Konsortialprojekt "B2B-RARE" zielt darauf ab, Diagnose- und Therapieverfahren für hereditäre NME zu entwickeln und die Patientenversorgung zu verbessern. Methoden: Hautbiopsien von 120 PatientInnen mit hereditären NME werden zur Fibroblastenisolation und multi-OMICs-Analyse entnommen. In ausgewählten Proben wird die Dichte intraepidermaler Nervenfasern untersucht. KI-gestützte Bioinformatik verknüpft die Ergebnisse mit Arzneimittel-Datenbanken, um potenzielle Therapien zu ermitteln, die nach Bestätigung in Muskel- oder Nervenzellen in Heilversuchen den Betroffenen zu Gute kommen. Zielsetzung: Das Projekt hat zum Ziel, personalisierte Therapiestrategien zu entwickeln, die direkt aus molekularen und zellulären Analysen abgeleitet werden. Diese Ansätze sollen die Behandlung von hereditären NME optimieren und auch Erkenntnisse für andere seltene Erkrankungen liefern. Schlussfolgerung: B2B-RARE könnte die Diagnostik und Therapie hereditärer neuromuskulärer Erkrankungen entscheidend verbessern. Minimal-invasive Hautbiopsien bieten eine innovative Diagnosetechnik, die neue Versorgungswege für seltene Erkrankungen eröffnen und die Lebensqualität der Betroffenen steigern soll.
