Abstract
Đặt vấn đề: Pazopanib hydroclorid là thuốc điều trị ung thư biểu mô tế bào thận và sarcoma mô mềm tiến triển, nhưng chưa có chuyên luận riêng trong các Dược điển tham chiếu. Tại Việt Nam, đã có nghiên cứu tổng hợp nhưng chưa có nghiên cứu về định lượng pazopanib hydroclorid. Vì vậy, cần xây dựng phương pháp định lượng pazopanib hydroclorid bằng sắc ký lỏng hiệu năng cao (HPLC) làm cơ sở xây dựng tiêu chuẩn chất lượng và đánh giá độ ổn định của sản phẩm sau tổng hợp. Mục tiêu nghiên cứu: Xây dựng và thẩm định phương pháp định lượng pazopanib hydroclorid bằng sắc ký lỏng hiệu năng cao. Đối tượng và phương pháp nghiên cứu: Chất thử là Pazopanib hydroclorid tổng hợp tại Trường Đại học Dược Hà Nội. Chất chuẩn: Pazopanib hydroclorid chuẩn của TRC, Canada. Thiết bị, dụng cụ: Hệ thống sắc ký lỏng hiệu năng cao Waters Acquity Arc (Mỹ), cân phân tích và các thiết bị khác đạt tiêu chuẩn phân tích. Phương pháp: Khảo sát điều kiện sắc ký đã công bố để xây dựng phương pháp phân tích phù hợp. Mẫu trắng là hỗn hợp acetonitril - nước (50:50), mẫu chuẩn và thử có nồng độ 500 µg/mL, lọc qua màng lọc 0,45 µL. Thẩm định phương pháp theo yêu cầu của ICH về các chỉ tiêu tính thích hợp hệ thống, độ đặc hiệu, độ chính xác, độ tuyến tính, độ đúng, độ lặp lại. Kết quả: Điều kiện sắc ký là cột C18 (250 mm x 4,6 mm; 5 µm), pha động pH 8 - acetonitril (55:45), tốc độ dòng 1,0 mL/phút, nhiệt độ cột 30 °C, detector DAD, bước sóng 269 nm, thể tích tiêm 10 µL. Phương pháp có độ đúng đạt yêu cầu (độ thu hồi từ 98,09 % đến 98,95 %), độ lặp lại tốt (RSD = 0,09 %), tương quan tuyến tính cao (r = 0,998) trong khoảng nồng độ 250-750 µg/mL. Kết luận: Phương pháp định lượng pazopanib hydroclorid bằng HPLC đã được xây dựng và thẩm định đạt.
