Cotton leaf curl Multan virus infecting <i>Acalypha wilkesiana</i> in China

Abstract
Cotton leaf curl Multan virus (CLCuMuV), a begomovirus originating in South Asia, has spread extensively across southern China and parts of Southeast Asia, primarily infecting Hibiscus rosa-sinensis. Here, we report the first identification of CLCuMuV and its associated betasatellite (CLCuMuB) in Acalypha wilkesiana in southern China. The virus (designated CLCuMuV-AW) and betasatellite (CLCuMuB-AW) isolated from A. wilkesiana share over 99% nucleotide identity with counterparts from H. rosa-sinensis in China but substantially lower identity with South Asian isolates. Phylogenetic analysis placed both CLCuMuV-AW and CLCuMuB-AW within a subclade composed exclusively of Chinese and Southeast Asian isolates, indicating a local origin from H. rosa-sinensis rather than a recent introduction. Notably, CLCuMuV-AW carries a unique 21-nucleotide deletion in its C4 open reading frame, representing a previously unreported genomic variant. Infectivity of CLCuMuV-AW was confirmed by agroinoculation of Nicotiana benthamiana using a tandem repeat construct. Overall, this study expands the known host range of the CLCuMuV–CLCuMuB complex and underscores the importance of continued monitoring.
